Product Description
This product is produced by immunizing rabbits with a synthetic lactylated peptide corresponding to residues surrounding Lys12 of human histone H4. Antibodies are purified by protein A-conjugated agarose followed by lactylated histone H4 (Lys12) peptide affinity chromatography.
Specificity
This antibody detects histone H4 only when it is lactylated at Lys12,but not the acetylated, propionylated, butyrylated, 3-hydroxybutyrylated, 2-hydroxyisobutyrylated or unmodified H4 peptide at Lys12.
Recommended Applications
ELISA, Dot blot, Western blot, Immunoprecipitation.

Important NOTES
For Western blotting, incubate membrane with diluted antibody in 5% nonfat milk, 1 x TBS, 0.1% Tween-20 for two hours at room temperature with gentle shaking.
Use at an assay dependent concentration. Optimal dilutions/concentrations should be determined by the end user.

Scientific Description
Histones are subjected to a variety of enzyme catalyzed modifications, including acetylation, methylation, phosphorylation,ubiquitylation, etc. The Warburg effect, which originally described increased production of lactate in cancer, is associated with diverse cellular processes such as angiogenesis, hypoxia, polarization of macrophages and activation of T cells. This phenomenon is intimately linked to several diseases including neoplasia, sepsis and autoimmune diseases.Lactate, which is converted from pyruvate in tumour cells, is widely known as an energy source and metabolic by-product. Zhang et al. (Nature, 2019) reveal that histone lactylation is a previously unknown histone modification,derived from the cellular metabolite lactate.
Storage & Stability:
The antibody is delivered in lyophilized powder. Upon receipt, please centrifuge the antibody at 12,000 x g for 20 seconds, and then reconstitute it with the antibody stabilizer provided. The stabilizer contains PBS, 50% glycerol, and 0.01% sodium azide. Please store the antibody at -20°C. Avoid repeated freeze/thaw cycles. Antibody is stable for 12 months from date of reconstitution. Equilibrate the antibody to room temperature for 2 minutes and mix by gently pipetting before use.
