Immunogen: The original mouse IgG1 version of this antibody was raised against sucrose-gradient purified monkeypox virus Copenhagen (MPXV Cph).
Specificity: This antibody recognizes monkeypox virus (MPXV). It also crossreacts with other orthopox viruses. Monkeypox virus (MPV) is a double-stranded DNA, zoonotic virus and a species of the genus Orthopoxvirus in the family Poxviridae. It causes a disease that has similar charactersists to smallpox but with a milder rash and a lower death rate. The virus causes disease in both humans and animals.
Application Notes: This antibody can be used to identify and analyse Monkypox virus (MPV). This clone was used to identify monkypox virus via ELISA (Czerny et al., 1994; pmid: 7513922). VACV-5C7 can be used to develop diagnostic assays which could allow for the detection of orthopox viruses, especially monkypox virus.
Storage Recommendation: Store at 4⁰C for up to 3 months. For longer storage, aliquot and store at - 20⁰C.

Immunogen: The original antibody was generated by immunizing BALB/c mice with WR strain of live vaccinia virus (VACV).
Specificity: This antibody binds the A13 membrane protein of the monkeypox virus and vaccinia virus. It recognizes an epitope comprising of a 10-amino acid sequence conserved in all orthopoxviruses, including viriola virus and monkeypox virus.
Application Notes: The original IgG2a version of this antibody bound to recombinant A13 protein expressed in E. coli, MBP conjugated VACV A13 in an ELISA. The original IgG2a antibody bound recombinant A13 proteins from E. coli with an affinity of 3.4 nM as detected by surface plasmon resonance. This antibody was also capable of recognizing a 12-kda protein in a Western blot of VACV-infected HeLa cells. This antibody stained viral factories and virion-size particles in immunofluorescence analysis of infected cells. This antibody was also reported to neutralized VACV mature virions in a plaque reduction assay. Passive immunization of mice with this antibody protected mice against intranasal VACV infection. This antibody generated against smallpox vaccine might contribute to immune protection against orthopoxviruses (PMID: 21810533).

Immunogen: The original mouse IgG2a version of this antibody was raised against sucrose-gradient purified Ectromelia virus (ECTV) Strain Munich 1.
Specificity: This antibody recognizes ECTV and some other orthopoxviruses. Its epitope is localized on a tetrapeptide located at the VACV homologue A27 (C-terminal epitope #5 (aa 68–71) (Ahsendorf et al., 2019; pmid: 31146446). Ectromelia virus (ECTV) is a virus of the family Poxviridae and the genus Orthopoxvirus that causes mousepox, a disease of mice. Mousepox causes skin lesions and generalized disease, which can be fatal. Orthopoxvirus has been associated with smallpox, cowpox, horsepox and monkeypox.
Application Notes: This antibody is recommended for the detection and analysis of Orthopox viruses, especially Ectromelia virus (ECTV). It binds to the epitope on the virus which seems not to be engaged with viral cell fusion (Ahsendorf et al., 2019; pmid: 31146446). VACV-5B1 (5B1/2G6) is a non-neutralizing clone that can be utilized for the detection of the analog of A27 protein on Orthopoxviruses (Ahsendorf et al., 2019; pmid: 31146446). This antibody's antigenic site is located in a functionally very important areawithin the C-terminus of the A27 protein; in this hydrophilic region, the two cysteines at positions71 and 72 are responsible for formation of disulfide bonds and, therefore, play an important rolefor a functionally active trimeric A27 structure (Ahsendorf et al., 2019; pmid: 31146446). Mouse IgG2a version of this clone (EM-5B1/2G6) was also used to develop an ELISA capture assay as a capture antibody and was shown to be superior in detecting Orthopox virues in comparison with other monoclonal and polyclonal clones (Johann and Czerny, 1993; pmid: 8122446). It makes it a very good candidate for the development of an anti-orthopox viruse detection assay.
