Description

The Antigen reactive T cell kit contains negative control (vial N) and positive control (vial P) in convenient packaging for stimulating human blood or PBMCs along with anti-human LFA-1 PE and surface stain cocktail consisting of anti-human CD45 PerCP, CD8 Pacific Blue™, CD4 FITC and CD14, CD41, CD15, CD16, CD19 APC/Cyanine7. This kit contains controls that can be paired with peptides of your choice for detecting antigen specific responses as well assessing vaccine responses. Cells or beads stained with individual fluorochrome-conjugated antibodies should be used for setting up compensation before running the samples. For data analysis, we recommend following gating strategy – Time –> Singlets –> Lymphocytes –> APC/Cyanine7 (Dump) negativeCD45+ –> CD4⁺CD8^– and CD8⁺CD4^–.

Formulation

Each kit will contain the following components:

Kit Components:

1 vial negative control
1 vial positive control
1 empty vial (for peptide stimulation)
1 vial surface stain cocktail (5 tests)
3 tubes of LFA-1 PE (1 test each) 1 bottle 50 mL RBC Lysis/Fixation solution
1 bottle 0.5 mL EDTA 0.08M
1 bottle 500 mL Cell Staining Buffer

Gating strategy using scatter parameters and surface markers in the staining cocktail to identify CD4+ and CD8+ cells.

CD8+ human peripheral blood lymphocytes were stimulated lyophilized negative control (left) or positive control (right) for 20 minutes, then stained with anti-human LFA-1 PE followed by surface cocktail. Data was analyzed using gating strategy depicted in Figure 3.
