potassium phosphate pH 조절 질문

transformation을 위해 버퍼를 만들고 있는데요.
논문에 나온 프로토콜대로 만들면 논문에서 제시한 pH가 맞지 않습니다.
transformation buffer는 다음과 같이 만듭니다.
Transformation buffer (2mM MgCl2, 5mM KH2PO4, 0.5M sucrose, 10% glycerol, pH 7.5) 200ml
MgCl2 (95.211 g/mol) 38mg and KH2PO4 (136.086 g/mol) 136mg (20ml)
dissolved in a small amount of DDW → add DDW to 20ml
Sterilized by 0.45μm sterile filter
Sucrose (342.3 g/mol) 34.23g (100ml)
dissolve it in a small amount of DDW by ultrasonication & agitation
→ add DDW to 100ml
Sterilized by 115℃, 20 min autoclaving → cool to room temperature
Glycerol 20ml (80ml)
dissolved in a small amount of DDW → add DDW to 80ml
Sterilized by 115℃, 20 min autoclaving → cool to room temperature
Mix all solutions in a clean bench after they are cooled to room temperature
프로토콜대로 다 만들고나면 보통 pH가 4.5에서 시작하는데요. 처음에는 1N NaOH를 1.8ml 정도 추가해서 pH를 7.5로 맞췄습니다.
그런데 그것 때문인지(는 잘 모르겠지만) transformation이 잘 안 됐습니다. 좀 더 찾아보니까 phostphate potassium buffer 같은 경우는 monobasic, dibasic을 섞어서 만들라고 하는 글을 봐서 계산 사이트에서 200ml(최종 부피) KH2PO4 몰 농도인 5mM을 맞춰놓고 pH 비율을 계산했는데 이렇게 하는 게 맞는 건가요? 논문에서는 KH2PO4 몰농도만 언급해서 K2HPO4를 함께 써도 되는지 모르겠습니다.
