sequencing에 사용하는 DNA pol.의 특성입니다.
따라서 vector에 있는 universal primer 위치가 insert 위치를 기준으로
상류, 하류로 약간 떨어져 있습니다.

왼쪽은 sanger sequencing의 앞부분이고, 오른쪽 그림은 끝부분입니다.
앞부분과 말단 부분의 sequencing 피크가 보통 이렇듯 불명확합니다.
그래서 피크가 명확하게 나오는 부분부터 시작해서 최대한 읽을 수 있는 끝부분까지만 잘라내서 sequencing 분석을 합니다.

DNA pol. 특징입니다.
sequencing mixture의 반응이 시작하면 primer + 1번 염기부터 정확히 합성하지
못합니다.
적어도 10개 base정도 지나면 정확히 base를 넣어줍니다.
그렇기 때문에 염기서열을 위한 primer는 분석하고자 하는 서열의 시작 서열 및
끝 서열을 기준으로 상, 하류로 거리를 두는 것이 원칙입니다.

conventional dye-labeled Sanger sequencing platforms perform poorly on short DNA fragments such as these when standard sequencing methodologies are applied. Specifically, these methods produce poor quality electropherograms at the 5′ end of the sequences, probably due to the irregular behavior and poor separation of the shortest DNA fragments during subsequent electrophoresis. Two methods have commonly been used to overcome this problem: (i) molecular cloning of the PCR products prior to sequencing, enabling sequencing primers located in the flanking vector DNA to increase the length and quality of the sequenced fragment and (ii) sequencing both strands in opposite directions, which produces two complementary sequences with good quality dual coverage in the middle and single coverage at the extremes.
