Adenovirus production and QC
재조합 adenovirus 제조의 경우 목적 유전자(GOI)를 운반하는 adenovirus 벡터는 우선 제한효소 PacI에 의해 선형화된다. 선형화된 벡터 DNA는 adenovirus 유전자 E1을 발현하는 packaging cell에 transfection되어 재조합 adenovirus를 생성하며, adenovirus 입자가 채취되는 조직 배양배지로 방출된다. Ultra-purified adenovirus (in vivo grade)의 경우 cesium chloride (CsCl) gradient ultracentrifugation에 의해 바이러스 입자가 더욱 정제되고 농축된다. 당사는 adenovirus-specific hexon protein을 검출하는 immunocytochemistry 기반의 방법을 사용하여 titer를 측정한다. Ultra-purified adenovirus의 경우 바이러스 입자의 optical density (OD260을 사용하여)를 측정하여 titer를 추산한다.
