1. 논문관련 분야의 소개, 동향, 전망을 설명, 연구과정에서 생긴 에피소드 영화나 게임에서나 볼 수 있었던 인공 생명체의 합성이 이제 인류의 눈 앞으로 조금씩 다가오고 있습니다. 십 수년 전 시작된 줄기세포에 대한 관심은 각 조직의 in vitro model, 즉 오가노이드(organoid) 연구로 이어졌습니다. 그리고 이제는 조직 뿐만 아니라 배아/태아의 in vitro model로서 실제 배아를 모방한 엠브리오이드(embryoid) 바람이 거세게 불고 있습니다. 그림1. 쥐 배아의 착상 및 발달과정 다들 한 번쯤 들어보셨을 배아줄기세포(embryonic stem cell, ESC)는 우리 몸의 모든 장기를 만들 수 있는 줄기세포로서, 착상 전 배아인 배반포(blastocyst)의 내부(inner cell mass, ICM)에서 얻을 수 있습니다 (그림 1). 우리 포유류는 타 동물종(species)과는 달리 모체 자궁 착상 그리고 태반을 통한 영양분 확보가 필수적입니다. 이를 위해 타 동물종에게는 없는 배아외조직(extraembryonic tissue)을 새로이 만들어 내게 되었고, 그것이 바로 영양막줄기세포(Trophoblast stem cell, TSC)입니다. 실제로 포유류의 영양막줄기세포는 발달 시기에 따라 여러 조직으로 분화하여 모체 자궁으로의 착상, 태아를 보호해 줄 탈락막 형성, 태반 형성을 조절하는 등 태아와 끊임없이 교류하며 태아 생존에 필수적인 역할을 합니다. 즉, 정교한 배아/태아 발달 연구를 위해서는 배아줄기세포 뿐만 아니라 영양막줄기세포 연구가 반드시 수반되어야 한다 는 뜻이죠. 하지만 안타깝게도 대부분의 배아 연구는 배아줄기세포에 초점이 맞추어져 있어 영양막줄기세포에 대한 정보가 많지 않고, 그나마 현재 널리 사용되는 유일한 영양막줄기세포주(TSC line)는 배반포 시기의 성격과 매우 달랐을뿐만 아니라 태반 특이적 마커를 보이는 등 이미 상당히 분화된 성질을 보여 주었습니다. 이는 제대로된 영양막줄기세포주 확보의 필요성을 뜻합니다. 분화되지 않은, 초기 배아의 영양막줄기세포를 완벽히 재현할 수 있는 세포주 확보를 위해 저는 ‘실제 배아에서는 한 쪽에 치우친 배아줄기세포의 위치 때문에 영양막세포의 운명이 결정된다’는 점에서 힌트를 얻고자 하였습니다. 쥐의 경우, 배아줄기세포와 먼 원거리 영양막세포(mural TSC)는 분화된 성격을 보이며 자궁착상을 유도합니다. 반면, 배아줄기세포와 가까운 근거리 영양막세포(polar TSC o
