1. 논문관련 분야의 소개, 동향, 전망을 설명, 연구과정에서 생긴 에피소드 자폐증(자폐 스펙트럼 장애)은 사회적 상호작용의 저하와 반복 행동의 증가로 진단되며, 유병률이 전 세계적으로 2.8%(36명 중 1명)에 달해 자폐증의 원인 규명과 치료법 발견이 절실합니다. 그동안 자폐증의 원인으로 유전적 요인 및 환경적 요인, 뇌의 기능 이상 등이 다양하게 보고되고 있으나, 아직까지 명확한 원인이 밝혀지지 않았습니다. 자폐증을 유발하는 것으로 보고된 1천개가 넘는 다양한 유전자 중에서도, 전사인자의 역할이 있는 ADNP는 높은 빈도로 자폐증을 유발하는 유전자로 알려져 있습니다. ADNP 유전자의 돌연변이는 자폐증 외에도 다양한 뇌 발달 장애를 유발하는데, 이를 통칭해 ADNP 증후군이라고 합니다. 본 연구에서는 ADNP 결손 생쥐 모델을 이용해 시냅스 능의 변화를 확인하고, 전사체 및 단백체 분석, 전기생리학적 분석 등으로 ADNP 결손 시 나타나는 자폐 관련 증상의 원인을 연구했습니다. 연구 결과, ADNP 결손 생쥐는 기억력 및 사회성 감소, 불안장애 증가 등 다양한 자폐 유사 증상을 보였으며, 기억력과 깊은 연관이 있는 시냅스 가소성의 일종인 시냅스 장기강화가 정상 생쥐에 비해 과도하게 증가되어 있는 것을 확인했습니다. 그리고 단백체 분석을 통해 칼슘/칼모듈린 의존 단백질 인산화효소(Ca2+/calmodulin-dependent protein kinase, CAMK)와 그 기질 단백질들의 인산화가 과도하게 증가된 것을 발견했습니다. 이러한 결과를 바탕으로, CAMK2 인산화 억제제(KN-62)를 처리했을 때 증가된 시냅스장기강화가 정상수준으로 회복되는 것을 확인했습니다. 이는 CAMK2의 과인산화가 ADNP 결손 생쥐에서 보이는 비정상적으로 증가된 시냅스 장기강화 및 자폐 관련 행동을 유발함을 나타내며, CAMK2의 활성 억제가 자폐 및 ADNP 증후군의 새로운 치료방법이 될 수 있음을 시사합니다. ADNP 결손 생쥐의 자폐증 유사 증상을 회복시키는 약물을 찾고자 오랜 기간이 걸려 포기하고 싶은 순간들이 많았는데, 다양한 공동연구자분들의 도움으로 연구를 성공적으로 마칠 수 있었습니다. 본 연구가 ADNP 증후군 및 자폐증 치료약 개발에 도움이 되길 바랍니다. 2. 연구를 진행했던 소속기관 또는 연구소에 대해 소개 부탁드립니다. 저는 KAIST 생명과학과 교수님이시자 IBS 시냅스 뇌질환 연구단 연구단장
