1. 논문관련 분야의 소개, 동향, 전망을 설명, 연구과정에서 생긴 에피소드 흔히 우울증 (major depressive disorder)을 "마음의 감기"라고 불리기도 하지만, 임상적인 우울증은 심각하고도 흔한 기분 장애 (mood disorder)로 몸과 마음을 동시에 황폐화시키는 심각한 질환입니다. 우울증은 치료받지 않으면 몇달 몇년에 걸쳐 지속되어, 인간관계의 와해나 직업적인 생산성의 상실, 자살에 이를 수도 있는 정신과 질환입니다. 인종에 관계없이 전세계적으로 10%정도의 사람들이 우울증을 앓고 있으며, 전체 인구의 20%정도가 평생동안 최소 한 번 정도의 임상적 우울증을 겪는것으로 알려져 있습니다. 한국에서도 OECD국가 중 자살율1위 그리고 자살 증가률 1위라는 부끄러운 두개의 기록을 가질 만큼 한국 사회에서도 우울증과 자살 문제가 심각한 사회문제로 떠오르고 있습니다. 현재 우울증의 원인은 두뇌에서 세로토닌 (serotonin), 도파민 (dopamine), 노르에피네프린 (norepinephrine)과 같은 대뇌 신경전달물질들의 생화학적인 불균형 (monoamine imbalance)이나, 부정적인 사고방식에 따른 심리학적인 불균형 (cognitive bias)에 기인하는 것으로 생각됩니다. 흔히 생화학적인 불균형과 심리학적인 불균형이, 외부의 스트레스 요인에 의해 촉발 또는 증폭되었을때 흔히 우울증이라고하는 심리적 유병사태에 이르는 것으로 생각되고 있습니다. 현재 우울증환자에게는 세로토닌 재흡수 억제제(Selective Serotonin Reuptake Inhibitor; SSRI)라고 하는 세로토닌계 약물요법이 가장 광범위하게 처방되고 있습니다. 그러나 이러한 세로토닌계 약물이 많은 우울증환자의 치료에 큰 도움이 됨에도 불구하고, 이상적인 약물 (ideal drug)이 되기에는 몇가지 큰 한계점들을 가지고 있습니다. 첫째로, 전체 투약군중에서 절반이 약간 넘는 60%정도의 환자에서만 효과가 있을뿐 나머지 30-40%정도의 환자에서는 큰 효험을 보이지 않는다는 점입니다. 불행히도, 이러한 치료저항성 환자들을 위한 대안 치료법 (alternative therapeutics) 또한 아직 미비한 상황입니다. 둘째로는 세로토닌계 약물은 중추신경계 전체 (central nervous system)에 퍼져 있는 세로토닌계 시냅스 (serotonergic synapse)에 광범위한 영향으로 인해, 투약 환자에 따라 불필요한 부작용을 수반하는 것으로 알려져 있습니다. 세째로, 세로토닌계 약물반응 (drug response)에 있

1. 논문관련 분야의 소개, 동향, 전망을 설명 Dr. Bliss와 그 동료들에 의해 시작된 후, 지난 30여년간 뇌의 기억 및 학습 기전연구에 중심이 되고 있는 것은 바로 공간지각능력이 있는 것으로 알려진 Hippocampus (해마)의 Synaptic Plasticity (신경연접가소성)에 대한 연구입니다. LTP (long-term potentiation)와 LTD (long-term depression)로 대표되는 Plasticity 현상은 활동의존적 조건변화에 따른 presynaptic neuron과 postsynaptic neuron 사이의 synaptic strength 변화입니다. 따라서, 오랜기간동안 glutamate에 반응하는 여러 receptor들의 trafficking이 연구되어 왔으나, dendrite와 spine들의 구조들이 상세히 밝혀지기 시작하면서 부분적으로 제한된 신경세포막 (활성화된 synapse나 주변 dendrite의 세포막)의 독립적 국소흥분성이 더불어 여러 연구팀들에 의해 관찰되고 있습니다. 세포막 흥분성의 조절은 주로 Na 2+ , Ca 2+ , K + 및 Cl - 의 ion channel들이 관여를 하고 있으며, 특히 postsynaptic neuron의 dendritic signal processing은 일차적으로 voltage-dependent A-type K + channel (transient outward K + current inducing channels, subtype Kv4.2 in CA1, Hoffman et al., 1997)에 의해 조절이 되고 있습니다 (ex, back-propagated Action Potential, EPSP amplification and transfer). 본 연구에서는 그동안 알려진 A-type K + channel의 dendritic membrane potential 조절기전과는 달리, 독립적으로 dendritic spine의 postsynaptic membrane potential에 관여하여 glutamate receptor의 하나인 NMDA receptor들의 자발활동성을 조절함으로써, synaptic sites의 Ca 2+ influx와 secondary signaling cascade에 영향을 미칠수 있다는 것을 최초로 보여준 것입니다. 특히 NMDA receptor들의 subunit composition 변화는 K + channel들이 synaptic plasticity (대부분이 NMDAR-dependence를 보이기 때문에)을 능동적으로 조절해줄 수 있다는 것입니다. 본 연구는 2007년에 본 연구실에서 발표한 논문 (Kim et al., 2007, Neuron)의 A-type K + channel들의 CA1 postsynaptic expression에 기초를 하였으며, 그에 따른 postsynaptic function에 촛점을 맞추었습니다. 현재 NMDA receptor들의 subunit composition은 brain synaptic development, synaptic Ca 2+ signaling, postsynaptic neuron에서의 Plas
