한빛사에 논문을 소개할 수 있게 되어 감사드립니다. 이번에 소개드리는 논문은 human genome 내의 대표적인 copy number variation (CNV) 인 amylase gene에 대한 분석을 수행한 논문으로 아래와 같은 네가지 정도의 핵심 내용을 소개하고 있습니다. Long-read sequencing 기술의 발달로 human genome을 완전히 읽어낼 수 있게 되면서 그동안 읽어낼 수 없었던 지역들에 대한 연구가 활발히 이루어지고 있습니다.

In this study, we overcome some of the traditional approaches to analyze the toxicity of nanomaterials that are used for various biomedical applications. In particular, many approaches on toxicity assessment of AgNPs have been previously studied using in vitro or in vivo analysis. However, there has been limited study on ex vivo toxicity evaluation, notably on human primary peripheral blood mononuclear cells (hPBMCs), which could be a feasible alternative approach to study leukocytes population.

Weight loss predicts faster disease progression in amyotrophic lateral sclerosis (ALS). Prior studies have focused on supplementing calories without identifying key macronutrients. In the current study, Dr. Ikjae Lee, in collaboration with Dr. Hiroshi Mitsumoto and colleagues from Columbia and the University of Kentucky, analyzed dietary patterns in the ALS COSMOS cohort to examine whether a specific macronutrient intake is associated with functional decline and survival.

췌장암은 5년 생존율이 12.5% 밖에 되지 않는 치사율이 매우 높은 암으로써, 90% 넘는 환자들에서 국소침범과 전이가 일어나는 것으로 알려져 있습니다. Next generation sequencing의 도입으로 췌장암의 carcinogenesis 에 관여하는 KRAS, CDKN2A, SMAD4, TP53 등의 cancer hotspot mutation의 역할에 대해서는 많이 알려져 있지만, 국소 침범과 전이의 기전에 대해서는 아직 많은 연구가 되어 있지 않습니다.

1. 논문관련 분야의 소개, 동향, 전망을 설명, 연구과정에서 생긴 에피소드 인체의 바이러스 감염 시에는 바이러스에 감염된 세포를 제거하는 기능을 가진 면역세포들이 중요한 역할을 하는데, 이들 중 NK 세포는 선천면역, T 세포는 적응면역의 특성을 보이는 대표적인 면역 세포입니다. 이 2가지 면역 세포는 바이러스에 감염된 세포를 인식하는 방식도 달라서, T 세포는 바이러스 단백질 조각을 항원으로 감지하는 반면 NK 세포는 스트레스 분자 발현이 증가한 것을 통해 바이러스 감염 세포를 감지합니다. 지금까지 면역학계에서는 이처럼 NK 세포와 T 세포가 명확히 구분되는 것으로 알려져 있었는데, 저희는 이번 연구를 통해 NK 세포와 T 세포의 특성을 모두 지니는 NK 유사 CD8 T 세포를 새롭게 발견하였습니다. 그리고 이러한 NK 유사 CD8 T 세포는 T 세포 수용체를 통해 바이러스 단백질 항원을 인식하는 대신에 NK 세포 수용체인 NKG2C를 통해 비정상 세포들을 인지하고 제거할 수 있다는 사실도 확인하였습니다. 이번 연구는, 그동안 면역학의 영역에서 이분법적으로 나누어져 있던 선천면역과 적응면역의 경계에서 작동하는 새로운 유형의 NK 유사 T 세포를 발견하고 그 특성을 밝힘으로써 인체의 면역 반응을 새로운 시각에서 바라볼 수 있게 하였다는 점에서 의의가 있습니다. 또한 저희 논문이 어셉트되기 직전에 스탠포드의 Mark Davis 그룹에서 또 다른 NK 수용체인 KIR 를 발현하는 CD8 T 세포가 과활성화 된 자가면역 CD4 T 세포의 기능을 억제할 수 있음을 Science 지에 발표하였고, 이러한 결과들을 토대로 저희 연구진은 NK 유사 CD8 T 세포가 체내에서 감염뿐만 아니라 각종 원인에 의해 비정상적으로 변한 세포들을 선택적으로 제거해 체내 항상성을 유지하는 데 중요한 역할을 한다는 가설을 가지고 후속 연구를 활발히 진행하고 있습니다. 2. 연구를 진행했던 소속기관 또는 연구소에 대해 소개 부탁드립니다. 제가 소속되었던 연구실에서는 암, 난치성 감염질환, 자가면역질환 등의 다양한 면역관련 질병을 대상으로 연구를 수행하고 있습니다. 구체적으로(1) 암환자의 종양 미세환경 내의 탈진화 세포와 면역 세포들의 특성 및 항종양 면역반응의 기전 연구, (2) 신/변종 바이러스 감염 질환에 대한 방어면역 특성 규명 및 백신 치료제 개발 연구, (3) 간질환(간암, 만성간염, 알콜성간질환, 간
