1. 논문관련 분야의 소개, 동향, 전망을 설명, 연구과정에서 생긴 에피소드 엽산 (folate)은 세포 분열에 필요한 핵산 (DNA와 RNA)의 합성을 촉매하는 필수 영양소입니다. 1948년 소아 병리학자였던 시드니 파버 (Sydney Farber)는 혈액암 세포의 증식과 엽산 대사 (folate metabolism)의 상관관계를 토대로 최초의 백혈병 치료제를 개발하였고, 이렇게 개발된 methotrexate 등의 엽산 회로 억제제는 오늘날에도 항암제 또는 면역 억제제로 널리 사용되고 있습니다. 세포 내 엽산 회로 (folate cycle)는 세포질과 미토콘드리아 경로로 구성됩니다. 유사한 화학반응을 촉매하는 효소들이 세포질과 미토콘드리아에 모두 존재하지만, 대부분의 암세포들은 미토콘드리아 경로를 이용하여 핵산 합성에 필요한 전구물질들을 생산한다는 것이 기존의 정설이었습니다. 따라서 지난 수년간, SHMT2 inhibitor와 같이 암세포 내 미토콘드리아 경로를 특이적으로 차단하는 항암제의 개발에 많은 노력이 기울여져 왔습니다. 하지만 미토콘드리아 엽산 경로는 암세포 뿐 아니라 생체 내 정상세포들도 활발히 이용하기 때문에, 미토콘드리아 경로의 억제는 정상세포에도 심각한 손상을 끼칠 수 밖에 없는 한계를 가지고 있었습니다. 한편 세포질 엽산 경로의 경우, 해당 경로의 핵심 효소 (SHMT1)가 유전적으로 제거되어도 정상세포의 생장과 발달에 크게 지장이 없다는 사실이 생쥐 실험을 통해 밝혀져 있었습니다. 우리 연구팀은 이러한 사실에 착안하여 세포질 엽산 경로에 의존하는 암세포들이 있는지, 암세포의 세포질 또는 미토콘드리아 경로 의존성을 조절하는 메커니즘은 무엇인지, 그리고 세포질 엽산 경로의 차단을 통해 특정 암세포들의 선택적인 성장 억제가 가능한지를 연구하였습니다. 결과적으로 본 연구는 대사체학 (metabolomics), 안정 동위원소 추적 (stable isotope tracing), 그리고 대사흐름분석 (metabolic flux analysis) 방법론들을 통해 (1) 상당수의 암세포들이 생리적 환경에서는 미토콘드리아 경로가 아닌 세포질 엽산 경로를 통해 핵산 합성에 필요한 전구물질들을 생산한다는 사실을 발견하였고, (2) 암세포 내 엽산의 농도가 암세포의 세포질 또는 미토콘드리아 경로 의존성을 결정한다는 사실을 밝혔습니다. 더불어 (3) 세포막에 존재하는 엽산 transporter인 SLC19A1의 발현이 세포 내 엽산 농도 조절에 핵심적인
