1. 논문관련 분야의 소개, 동향, 전망을 설명, 연구과정에서 생긴 에피소드 대장암은 세계적으로 가장 빈번하게 발생하는 암 중 하나입니다. 대장암의 발병인자로서 동물성 지방의 위험성이 알려져 있습니다. 담즙산은 간에서 콜레스테롤로부터 생성되어 1차 담즙산 (Primary bile acids)의 형태로 소화기계로 분비 되며, 음식물로 섭취한 지방질의 흡수에 관여합니다. 대부분의 담즙산은 소장에서 대부분 재흡수되지만, 고 지방식을 하는 경우 대장에서 담즙산의 축적이 발생하게 되고 장관 내 혐기성 세균에 의해서 1차 담즙산에서 2차 담즙산 (Secondary bile acids)으로의 전환이 이루어집니다. 디옥시콜릭산 (DCA: deoxycholic acid) 과 리토콜릭산 (LCA: lithocholic acid) 과 같은 2차 담즙산은 세포에 대한 독성을 유발하여 장간 순환 과정에서 장내 상피세포를 손상시키고 손상된 세포의 자멸사를 억제하여 대장암의 발병 기전에 관여하는 것으로 알려져 있습니다. 인간의 간은 콜레스테롤로부터 주로 sterol 고리가 “Kinked: 꼬인“ 형태의 담즙산을 생성하지만, sterol 고리가 “Flat: 평평한” 형태의 담즙산도 소량으로 생성합니다. 복잡한 다단계 담즙산 7α-탈수소화 경로를 통해 콜릭산 (Cholic acid, CA) 과 키노디옥시콜릭산 (Chenodeoxycholic acid, CDCA)과 같은 “Kinked” 1차 담즙산의 디옥시콜릭산 (Deoxycholic acid, DCA) 및 리토콜릭산 (Lithocholic acid, LCA)과 같은 “Kinked” 2차 담즙산으로의 효소 전환은 비교적 잘 연구 되어 있습니다. 그러나, 장내 미생물군 피르미큐테스 (Firmicutes)에 의한 “Kinked” 1차 담즙산으로 부터 알로디옥시콜릭산 (allo-deoxycholic acid, allo-DCA) 및 알로리토콜릭산 (allo-lithocholic acid, allo-LCA)과 같은 “Flat” 2차 담즙산으로의 효소 전환은 약 30여년동안 잘 알려져 있지 않았습니다. 본 연구에서는 피르미큐테스에서 BaiP 와 BaiJ 단백질이 “Flat” 2차 담즙산 allo-DCA 및 allo-LCA 생성에 관여하는 것을 규명하였습니다. 선행연구에 따르면 2차 담즙산의 미세한 화학적 구조의 차이 (Kinked, Flat) 에 따라 세포 독성 및 생물학적 영향들이 상이한 것으로 현재 예상하고 있습니다. 그 예로, 최근 일본에서 장내 담즙산의 특이적인 프로파일이 일본의 100세인들이 건강하게 늙는 원인일 가능성이 높다는 연구 결과를 발표하였습니다. 일반적으로 2차
