1. 논문관련 분야의 소개, 동향, 전망을 설명, 연구과정에서 생긴 에피소드 Organelle contacts 혹은 Membrane contact sites 관련 연구는 최근 새롭게 주목받고 있는 분야로, 관련 연구자들 사이에서조차 통일된 용어가 없을 만큼 이제 막 시작하는 단계에 있습니다. 제가 처음 박사과정을 시작하던 당시 저희 랩에서 처음으로 human cell line에서 Mitochondria-lysosome contacts (Wong et al. Nature, 2018)의 형성, 기능 및 기전을 발표하였고, 이후 현재까지 저희 랩에서 발표한 3건의 후속연구를 포함하여 총 9~10 편의 후속 연구들이 발표되었습니다. 이번 Nature communications 논문은 특히 iPSC 분화를 통해 만들어진 도파민 뉴런에서 Mitochondria-lysosome contact dynamics를 보는데 중점을 두었는데, 특히 mitochondrial dysfunction과 lysosome dysfunction이 모두 파킨슨병의 기전으로 알려져 있다는 사실에 기반하여 파킨슨병 환자의 뉴런에서 손상된 Mitochondria-lysosome contact가 질병의 진행에 어떠한 영향을 줄 수 있는지를 확인했습니다. 기존에는 단일 세포소기관 수준에서 연구되던 질병의 기전들이, 여러 다른 세포소기관들의 상호작용 수준에서 새로이 주목받고 있고, 특히 알츠하이머, ALS 등 신경퇴행성질환에서 ER-mitochondria contacts 등 세포 소기관 간의 상호작용이 중요한 역할을 한다는 사실이 하나 둘 보고되고 있습니다. 세포 소기관 간의 상호작용을 연구하는 저에게는 이렇듯 점차 확장되어가는 필드에 대한 관심을 지켜보는 일이 큰 즐거움입니다. 모든 연구가 저마다의 어려움을 가지고 있을 텐데요, 제가 연구를 진행하는 과정에서 겪었던 가장 큰 어려움은 실험 모델과 관련된 부분이었던 것 같습니다. 최소 50일 길게는 300일까지 배양을 해야 재료로 사용할 수 있는 iPSC-derived neuron의 특성상 실험 재료 준비에만 몇 달이 걸려 좋은 아이디어가 있어도 그 아이디어가 데이터로 보여지기까지 한 호흡이 너무 길었고, 6개월 이상 배양한 뉴런을 이용한 실험에서 아무런 결과를 얻지 못하는 날이면, 연구 전반에 대한 회의감에 힘든 시간들을 보내야 했습니다. 다행스럽게도, 현미경 아래 살아 꿈틀대는 뉴런들을 이미징 하는 일은 항상 가슴 설레는 일이었고 어느덧 Cell Mom 역할이 저의 천직인 양 익숙해져 가는 느낌입니다. 짧게, 저희 교수님 자랑으로 인터뷰 첫 질문
