1. 논문관련 분야의 소개, 동향, 전망을 설명, 연구과정에서 생긴 에피소드 Lineage tracing provides information about the number of progenies of the founder cell, their growing pattern and differentiation status. Recently, many lineage tracing approaches have been developed to track a cellular lineage in diverse cells, tissues and whole organisms. The early processes in embryogenesis have been studied in the model organisms using direct observation. The complete developmental cellular lineage of C. elegans was reconstructed by using the microscopic observation, leading this study awarded the Nobel Prize in 2002 (Sulston et al. 1983, Dev Biol). After then, many researchers have used direct observation approach for studying cellular lineages during embryogenesis in zebrafish (Kimmel CB and Warga RM. 1986, Science) and fruit-fly (Campos-Ortega and Hartenstein. 1985, Springer-Verlag Berlin Heidelberg) by developing different microscopic techniques. Recently, genetically modified mice (such as Brainbow mice and Confetti mice etc.) have also become the mainstream of genetic lineage tracing studies, using Cre-loxP and FLP-FRT systems (Livet et al. 2007, Nature; Snippert et al. 2010, Cell). These technologies have been used for the expression of the fluorescent proteins or polylox cassette (excisions and inversions), also used as genetic barcode to study the clonal output, cellular diversity and tissues during embryonic development, cell differentiation and tissue regeneration (Pei et al. 2017, Nature). In addition, CRISPR-Cas9 barcode editing approach with single cell sequencing and genome editing of synthetic target arrays for lineage tracing (GESTALT) technique in animal models (Yang et al. 2014, Nat Commun; McKenna et al. 2016, Science) have been developed for elucidating accurate lineage trees for the early embryonic developmental stages.
