  1. 논문관련 분야의 소개, 동향, 전망을 설명, 연구과정에서 생긴 에피소드 Alpha-helical membrane protein의 folding에 대한 연구에서 가장 중요하고도 어려운 문제는 membrane protein들이 lipid bilayer에서 가역적 (reversible)으로 folding하고 assembly할 수 있는 시스템을 디자인하는 일입니다 (Bowie, 2005). Water-soluble protein의 경우 reversible folding이 매우 명확합니다 (thermodynamic control) (Baker and Agard, 1994). 의아하게 생각하실지 모르겠지만 membrane protein의 경우에는 detergent micelle 상에서의 reversible folding 시스템이 개발되었지만 lipid bilayer상에서는 아직까지 그러한 예가 없습니다. 또한 membrane protein folding이 대부분의 water-soluble protein의 경우와 같이 thermodynamic control에 의해 지배되는지에 대해서는 아직 실험적인 정보부족으로 이분야의 과학자들이 선뜻 동의하지 못하고 있습니다. Membrane protein의 X-ray 구조가 1984년에 처음 발표되었고 (Deisenhofer et al., 1984), 곧 이어 Popot과 Engelman에 의해 alpha-helical membrane protein folding의 모델인 “two-stage model”이 처음 제시되었지만 (Popot et al., 1987) folding의 driving force나 메카니즘에 관한 연구는 실제 membrane protein이 존재하는 lipid bilayer에서의 연구의 어려움 때문에 water-soluble protein에 비해 매우 더디게 발전되어 왔습니다. 그럼에도 불구하고, Beta-barrel membrane protein의 경우에 몇몇 bacterial outer membrane protein들에서 reversible folding 시스템이 개발된 예가 있습니다 (Hong and Tamm, 2004; Huysmans et al.). 본 논문에서 저희들이 가졌던 가장 큰 문제의식은 lipid bilayer에서 강하게 상호작용하는 alpha-helical membrane protein assembly 간의 affinity (thermodynamic stability)를 어떻게 정량적으로 측정할 수 있을까하는 것이었습니다. Lipid bilayer에서의 단백질간의 상호작용은 membrane protein의 구조와 기능을 이루게하는 folding과 assembly, 그리고 많은 signaling pathway에서 매우 중요합니다 (Matthews et al., 2006; Moore et al., 2008). 어떻게 보면 단백질간의 affinity를 측정하는 것은 생화학에서 가장 기본적인 일 중의 하나입니다만, membrane protein간의 상호작용을 intact lipid bilayer에서 reversible 시스템
