1. 논문관련 분야의 소개, 동향, 전망을 설명, 연구과정에서 생긴 에피소드 예로부터 박테리아 감염은 인간에게 위협이 되었습니다. 그 중에도 그람음성균에 감염되면 그 외막 구성성분인 LPS를 우리 몸의 선천적 면역 시스템에서 중요한 TLR4가 인식하여 면역반응이 일어나게 만드는데 이러한 면역반응이 과도해질 경우 사이토카인 폭풍이 유도되어 인간에게 치명적인 패혈증을 유발하게 됩니다. 이러한 감염증은 항생제의 등장으로 정복되는 줄 알았으나 항생제 내성균주의 출현은 항생제의 한계를 보여주며 새로운 작용기전의 항균물질의 필요성이 대두되었고 대체제로 등장한 것 중 하나가 세균의 세포막을 타겟으로 하는 항균펩타이드 (Antimicrobial peptide)입니다. 저희는 그 중 곤충의 세크로핀 계열 항균펩타이드 Papiliocin이 그람음성내성균 치료의 최후의 보루로 불리는 Polymyxin보다도 LPS 제거에 더 효과적이라는 사실을 발견하였습니다. 저희는 Papiliocin이 LPS를 제거할 뿐 아니라 LPS와 유사한 결합 패턴으로 LPS와 TLR4의 결합을 방해함으로써 과도한 면역반응을 제어해 패혈증을 원천적으로 치료하는 핵심원리를 구조 기반 상호작용으로 규명하였습니다. 또한 패혈증 동물모델에서 Papiliocin의 치료효과를 확인함으로서 앞으로 패혈증 치료제 개발의 기반을 마련하였습니다. 2. 연구를 진행했던 소속기관 또는 연구소에 대해 소개 부탁드립니다. 건국대학교 일반대학원 생명공학과 구조화학연구실 (Konkuk university, Structural chemistry lab)에서 김양미 교수님 지도하에 석사과정을 마쳤습니다. 저희 연구실은 NMR, x-ray, 분자모델링을 이용하여 단백질과 항균펩타이드의 삼차원 구조를 연구하고, 구조-활성 상관관계에 기반하여 항생제를 개발하고 있습니다. 연구실에서는 패혈증 치료제 개발을 목표로 연구를 수행하였으며, 동물 실험실을 비롯한 다양한 실험 시설을 갖추고 있기에 대부분의 연구를 직접 수행할 수 있었고 추가적인 실험이 필요한 경우에는 한국기초과학지원연구원의 장비를 이용하여 연구를 수행하였습니다. 3. 연구 활동 하시면서 평소 느끼신 점 또는 자부심, 보람 석사학위 과정 동안 다양한 실험을 진행해보았고 수차례에 걸친 실패와 논의를 통해 연구의 어려움을 알게 되었지만 그 성과에 대한 성취감도 느낄 수 있었습니다. 특히 제가 대학원 진학을 결심할 때
