2023년 9월부터 단국대학교에서 독립된 ‘영양세포대사생화학 연구실 (Lab of Nutritional and Cellular Metabolism & Biochemistry,
https://cmbsince2023.imweb.me/)’을 운영하며, 영양소들의 세포내 대사 과정에서 생성이 되는 중간 대사체들의 비정형적인 역할을 규명하는 연구를 진행해 오고 있습니다.
특히, 1) 필수 영양소 (예, 셀레늄) 유래 중간대사물의 새로운 역할을 규명하고, 2) 영양소 유래 기능성 대사체들이 어떻게 세포의 삶과 죽음을 조절하는지 초점을 맞추어 연구를 해오고 있으며, 특히 페롭토시스를 조절하는 대사체 및 대사경로를 연구하고 있습니다. 이를 바탕으로 암, 선천성대사질환 및 대사성 관련 질환의 치료방법을 모색하고자 합니다. 장기적으로는 3) 각 영양소 대사의 전신 생체지도를 구축하고 각 개체의 개별 맞춤 영양 전략 및 질병치료에 활용하고자 합니다. 이러한 연구를 위해서 대사체 분석 및 CRISPR pooled 스크리닝 등 다양한 대사 연구 최신 연구 기법을 활용하고 있습니다.
What we are interested
The Cellular Metabolism and Biochemistry Laboratory focuses on investigating the novel roles of metabolic intermediates generated within cells. In particular, our research aims to understand how these intermediates regulate cell survival and death. Ultimately, we strive to leverage these findings to develop therapeutic strategies for cancer, inborn errors of metabolism, and other metabolic-related diseases. To achieve these goals, we employ cutting-edge research techniques such as metabolomics analysis and CRISPR pooled screening. Our specific research topics and objectives are outlined below.
1. Exploring Novel Roles of Selenium Metabolism
Recent advancements in metabolomics have revealed that known nutrients often possess functions beyond their traditional roles within cells. Our recent study demonstrated that hydrogen selenide, a metabolic intermediate in selenocysteine biosynthesis, is utilized by the sulfide quinone oxidoreductase (SQOR) enzyme to reduce coenzyme Q10, thereby protecting cells from ferroptosis. Building on this finding, we are actively investigating additional novel functions of selenium metabolism to uncover its broader biological significance.
2. Exploring Non-Canonical Roles of Metabolic Intermediates in Cancer
Cells generate various metabolic intermediates, traditionally viewed as precursors for energy production, building blocks, or redox regulation. However, emerging research suggests that these intermediates possess distinct and independent biological functions. Our lab is focused on identifying these non-canonical roles of metabolic intermediates and exploring their therapeutic potential. By leveraging these findings, we aim to develop innovative strategies for treating cancer using cellular metabolites as therapeutic agents.
3. Elucidating the Toxicity Mechanisms of Metabolites in IEM and Related Diseases
Inborn errors of metabolism (IEM) are caused by mutations in metabolic enzymes, leading to the accumulation of toxic metabolites rather than a deficiency in their products. These disorders, with over 500 types identified, often lack effective treatment options. Despite their clinical importance, appropriate experimental models for studying IEM remain underdeveloped. Our research aims to establish robust disease models to investigate the mechanisms of metabolite-induced toxicity and to explore novel therapeutic interventions for these metabolic disorders.
4. Exploring Metabolic Pathways Regulating Ferroptosis
We recently identified SQOR as a novel anti-ferroptosis regulator. Ferroptosis, an iron-dependent form of cell death, is regulated by diverse metabolic pathways and is implicated in diseases ranging from cancer to neurodegeneration. Our lab aims to uncover additional metabolic pathways regulating ferroptosis, with the ultimate goal of translating these findings into therapeutic strategies for ferroptosis-associated diseases.
These research endeavors collectively aim to advance our understanding of cellular metabolism and its implications for human health and disease.
