Abstract
Owing to the increased antimicrobial resistance, a new therapeutic platform is urgently needed to combat microbial infections with therapies other than traditional antibiotics. We developed a zeolitic imidazolate framework (ZIF8) nanocarrier-based nanotherapy for the natural substances berberine (BER) and/or carvacrol (CAR), composed of a chlorin e6 photosensitizer (Ce6), to achieve phototherapy. The resulting nanoformulations, namely ZIF8-CAR/Ce6, ZIF8-BER/Ce6, and ZIF8-CAR-BER/Ce6, were produced and tested against S. aureus and P. aeruginosa in vitro. The nanoformulations size varied between 200 and 255 nm, with high entrapment efficiency and drug-loading content. Controlled, pH-responsive release of CAR and BER from ZIF8 was observed, with significantly enhanced release at pH 5.5 and 7.4. S. aureus exhibited a primary singular killing effect attributable to reactive oxygen species (ROS) generated by the nanoformulation, whereas P. aeruginosa exhibited a simultaneous killing effect from ROS and heat under 808-nm laser irradiation. This demonstrates the selective killing and synergistic efficacy of a single 808-nm laser. The superior bactericidal effects induced by the nanoformulations were validated through various physiological characteristics, including ROS production, cell morphology, membrane potential, biofilm, and hemolysis effect. These findings underscore the potential of integrating natural substances with phototherapy in nanotherapy as a promising antibacterial therapy against drug-resistant bacteria.
