Abstract

Amyloid-β (Aβ) oligomers are implicated in the onset of Alzheimer's disease (AD). Herein, quinoline-derived half-curcumin-dioxaborine (Q-OB) fluorescent probe was designed for detecting Aβ oligomers by finely tailoring the hydrophobicity of the biannulate donor motifs in donor-π-acceptor structure. Q-OB shows a great sensing potency in dynamically monitoring oligomerization of Aβ during amyloid fibrillogenesis in vitro. In addition, we applied this strategy to fluorometrically analyze Aβ self-assembly kinetics in the cerebrospinal fluids (CSF) of AD patients. The fluorescence intensity of Q-OB in AD patients' CSF revealed a marked change of log (I/I 0) value of 0.34 ± 0.13 (cognitive normal), 0.15 ± 0.12 (mild cognitive impairment), and 0.14 ± 0.10 (AD dementia), guiding to distinguish a state of AD continuum for early diagnosis of AD. These studies demonstrate the potential of our approach can expand the currently available preclinical diagnostic platform for the early stages of AD, aiding in the disruption of pathological progression and the development of appropriate treatment strategies.

알츠하이머병 환자 뇌척수액 내 Aβ 올리고머 감응 형광체를 활용한 조기진단 플랫폼 개발 [Nat. Commun.] 아밀로이드 베타(Amyloid β, Aβ) 올리고머는 알츠하이머병(Alzheimer’s disease)의 주요한 바이오 마커이다. 본 연구에서는 Aβ 올리고머의 선택적 검출을 위한 퀴놀린(Quinoline)과 디옥사보린(Dioxaborine) 구조가 결합된 커큐민(Curcumin) 유도체 저분자 형광 프로브(Q-OB)를 개발하였다. Q-OB는 용액상에서 Aβ 섬유화(Fibrogenesis) 과정 중 Aβ 올리고머화(Oligomerization)를 동적으로 모니터링할 수 있는 분광학적 특성을 보였다. Q-OB를 활용하여 알츠하이머병 환자의 뇌척수액 내에서 Aβ의 자가 응집 속도를 형광 분석하였으며, 이를 통해 알츠하이머병 환자와 정상 대조군의 비교에서 조기 진단의 지표로 활용할 수 있는 유의미한 측정값을 얻었다. 그 결과, 해당 연구는 현재 상용화된 임상 진단 플랫폼을 확장하여 알츠하이머병의 조기진단에 활용될 수 있는 체외진단 플랫폼 개발의 가능성을 제시했다.
