In nature there are various compounds which elicit the taste sensation in animals. These include macronutrients, such as carbohydrates, lipids, proteins, and micronutrients, such as minerals. Although vitamins are typically thought of as micronutrients, it is unknown how they are perceived. Thus, in our study entitled “The Taste of Vitamin C in Drosophila”, we endeavored ourselves to identify the molecular and cellular mechanism of vitamin C perception using the highly tractable model organism Drosophila melanogaster. First, we demonstrated that vitamin C can enhance the survival in starvation condition and enhance the fecundity of animal. Next, we identified that the fly prefers the optimum concentration of vitamin C via sweet-sensing gustatory receptor neurons (GRNs) in the labellum, a external taste organ of fly. Further, we also revealed that fly can sense vitamin C via highly divergent subfamily of ionotropic glutamate receptors (iGluR) called ionotropic receptors (Irs) along with gustatory receptors (Grs). More concisely, we found the role of broadly tuned IR25a and IR76b along with GR5a, GR61a, GR64b, GR64c, and GR64e in sensation of vitamin C. For identifying the above-mentioned molecular sensors, we utilized feeding assay, oviposition assay and electrophysiology (tip recording assay). Our conceptual presentation and critical analysis in this study can provide insight into how optimum consumption of micronutrients such as vitamins is advantageous to animals and what molecular sensors are required to detect them.
The authors of this paper are affiliated with the Department of Bio & Fermentation Convergence Technology at Kookmin University, Seoul. In our molecular genetic lab, we analyze taste sensation, cellular neurobiology of temperature sensation, circadian rhythm, sleep regulation, and metabolism. We employ behavioral assay, electrophysiology, immunohistochemistry, DrosoX technique, metabolic measurement assay and other molecular techniques. By studying above mentioned area, we aim to explain the detailed mechanism of chemosensation, circadian rhythm & metabolism of animal and contribute to scientific community for better understanding of socio-physiological behavior of animal.
I feel that while conducting research, I feel a sense of pride in my work as it contributes new knowledge to the scientific community. Even my research leads to breakthroughs or advancements with a significant impact on society. Besides, the research activities are challenging which intellectually stimulate me by allowing me to stretch my skills and knowledge in new ways. This is a kind of accomplishment and personal growth. I am very oblige to Prof. Youngseok Lee for providing me platform to perform and share my research with others, such as presenting at conferences or publishing papers. Also, a supportive work environment and lab colleagues who have my best interests in mind are a great source of motivation.
As many people are interested in research activities, I would suggest that if they are interested in pursuing research in molecular genetics, that's great! This is an exciting field with many opportunities for discovery and innovation. I believe that they can have a great future and can provide valuable feedback in the academic community. Just be persistent and resilient as research can be challenging and setbacks are common. Don't be discouraged if experiments don't work or if you encounter obstacles along the way. Learn from your mistakes and keep moving forward. I would also suggest building a strong network of mentors and colleagues. Collaborate with other researchers, seek out advice and feedback from mentors, and attend conferences and networking events to meet others in the field. This will lead you to achieve your goal. Remember, research is a journey, and the most important thing is to stay curious, passionate, and committed to your work.
As research is never ending process, I am further planning to enhance my research activities to a higher level. The skills I learned through my PhD life have prepared me well to transition from studying peripheral neuronal response analysis to higher order neuronal activities. Additionally, I look forward on applying my molecular genetic skills towards brain connectome analysis examining the effect of higher order neuronal circuits in neurodegenerative disease and their pattern of projections leading to connector circuits damage. I wish to accomplish these achievements for which I am preparing myself in the present scenario.
I would like to take a moment to express my sincere gratitude to Prof. Youngseok Lee for his invaluable guidance and support throughout this study. His expertise, feedback, and encouragement have been instrumental in helping me navigate the challenges of research and achieve my goals. I would also like to thank my present and past lab members, my family in Nepal, and my charming wife for their unwavering commitment and for inspiring me to do my best work. Finally, I would like to thank the Kookmin University family for providing me with the opportunity to join them.
