pluriMate Separation Tubes 는
밀도 구배 원심분리(density gradient centrifugation)를 기반으로 전혈(whole blood) 및 골수(bone marrow) 시료로부터 백혈구(leukocytes), 백혈구 아형(림프구 및 단핵구), 말초혈액 단핵세포(PBMC), 혈소판을 신속하고 효율적으로 분리·농축할 수 있도록 설계된 전용 분리 튜브입니다.

튜브 내부의 barrier 구조를 통해 시료 주입 시 밀도 구배 배지(density gradient medium)와 시료가 섞이는 것을 최소화하도록 설계되어 있습니다. 이를 통해 작업자는 시료를 분리 배지 위에 조심스럽게 적층해야 하는 기존 방식의 불편함을 해소하여, 보다 쉽고 빠르게 주입할 수 있으며, 원심분리 후에는 목표 세포층을 보다 간편하게 회수할 수 있습니다.

또한 Leucosep 또는 SepMate 튜브를 대체할 수 있는 실용적인 대안으로 활용할 수 있습니다.

pluriMate 분리 원리

밀도 구배 원심분리법은 원심분리와 일정한 밀도로 조정된 분리 매질을 결합하여 세포를 분리하는 방법입니다.

pluriMate 튜브 내부에 밀도 구배 배지 density gradient medium 와 시료를 넣고 원심분리를 진행하면, 각 세포는 밀도 차이에 따라 서로 다른 층으로 분리됩니다.
이때 목표 세포는 지정된 분리층에 집적되며, 원하지 않는 세포들은 하층으로 이동하게 됩니다.
이 구조를 통해 목표 세포를 보다 쉽고 정확하게 회수할 수 있습니다.

원심분리 전 튜브 층 분포

먼저 pluriMate 튜브에 적절한 밀도 구배 배지 density gradient medium(3)를 pluriMate barrier(2) 높이까지 채운 후, 시료(예: 전혈)를 튜브에 붓거나 피펫으로 분주합니다

이때 내장된 pluriMate barrier(2)가 시료 주입 시 배지(3)와 시료(1)의 혼합을 방지해줍니다.
따라서 시료를 빠르게 처리해야 하는 경우, 시료를 튜브에 그대로 부어 넣는 것만으로도 간편하게 작업할 수 있습니다.

원심분리 후 튜브 층 분포

원심분리가 완료되면 튜브 내부의 층 구조가 변화합니다.
pluriMate barrier(4) 위쪽에서 Plasma (1)와 밀도 구배 배지(3) 사이에 **목표 세포-Leukocytes(2)**가 위치하게 됩니다.

반면 pluriMate barrier(4) 아래쪽에는 일부 밀도 구배 배지(3)와 함께, 적혈구, 혈소판 및 사멸 세포(dead cells) 등 원하지 않는 세포들(5)이 분리되어 남게 됩니다.
